Mendelian Randomization in Cardiovascular Research

چکیده

Mendelian randomization is an epidemiological approach to making causal inferences using observational data. It makes use of the natural that occurs in generation individual’s genetic makeup a way analogous study design randomized controlled trial and uses instrumental variable analysis where variant(s) are instrument (analogous random allocation treatment group trial). As with any variable, there 3 assumptions must be made about instrument: (1) it associated (not necessarily causally) exposure (relevance condition); (2) outcome only through (exclusion restriction (3) does not share common cause (ie, no confounders outcome, independence condition). Using example type II diabetes coronary artery disease, we demonstrate how method may used investigate causality discuss potential benefits pitfalls. We conclude although studies can usually establish on their own, they usefully contribute evidence base increase our certainty effectiveness (or otherwise) interventions reduce cardiovascular disease.

Mendelian Randomization Causal Analysis Adiposity as a cause of cardiovascular disease: a Mendelian randomization study

Background: Adiposity, as indicated by body mass index (BMI), has been associated with risk of cardiovascular diseases in epidemiological studies. We aimed to investigate if these associations are causal, using Mendelian randomization (MR) methods. Methods: The associations of BMI with cardiovascular outcomes [coronary heart disease (CHD), heart failure and ischaemic stroke], and associations o...

Moderate Alcohol Use and Cardiovascular Disease from Mendelian Randomization

BACKGROUND Observational studies show moderate alcohol use negatively associated with ischemic heart disease (IHD) and cardiovascular disease (CVD). However, healthier attributes among moderate users compared to never users may confound the apparent association. A potentially less biased way to examine the association is Mendelian randomization, using alcohol metabolizing genes which influence ...

Cystatin C and Cardiovascular Disease A Mendelian Randomization Study

BACKGROUND Epidemiological studies show that high circulating cystatin C is associated with risk of cardiovascular disease (CVD), independent of creatinine-based renal function measurements. It is unclear whether this relationship is causal, arises from residual confounding, and/or is a consequence of reverse causation. OBJECTIVES The aim of this study was to use Mendelian randomization to inve...

Uric Acid and Cardiovascular Events: A Mendelian Randomization Study.

Obesity and diets rich in uric acid-raising components appear to account for the increased prevalence of hyperuricemia in Westernized populations. Prevalence rates of hypertension, diabetes mellitus, CKD, and cardiovascular disease are also increasing. We used Mendelian randomization to examine whether uric acid is an independent and causal cardiovascular risk factor. Serum uric acid was measur...

5. Mendelian randomization

Associations between modifiable exposures and disease seen in observational epidemiology are sometimes confounded and thus misleading, despite our best efforts to improve the design and analysis of studies. Mendelian randomization, the random assortment of genes from parents to offspring that occurs during gamete formation and conception provides one method for assessing the causal nature of so...
